Анотація
Проведено дослідження виділених з Чорного моря спороутворюючих бактерій щодо здатності до деградації ситетичих полімерів. Динаміку активності визначали методом скринінгу на агарових пластинах з додаванням тестових сполук. У геномах найбільш активних штамів проведено пошук за допомогою прихованої марковської моделі з метою виділення послідовностей ферментів, відповідальних за деградацію.
Виявлено 52 культури, здатні до розкладання синтетичних полімерів in vitro. Найвищу розкладаючу активність щодо імпранілу показали штами B. velezensis та B. halotolerans (B103), щодо БГЕТ – два штами B. atrophaeus (B82, B86). Установлено, що розкладання імпранілу активніше відбувалося при 30 ℃, на розкладання біс(2-гідроксіетил)терефталату впливу температури не зафіксовано. У геномах активних культур виявлено 45 потенційних ПЕТаз, що належать до надродини α/β гідролаз.
A study was conducted on the ability of spore-forming bacteria isolated from the Black Sea to degrade synthetic polymers. The dynamics of activity was determined by screening on agar plates with the addition of test compounds. The genomes of the most active strains were searched using hidden Markov model in order to isolate the sequences of enzymes responsible for degradation. A total of 52 cultures capable of degrading synthetic polymers in vitro were identified. The highest degradative activity toward Impranil was demonstrated by the strains B. velezensis and B. halotolerans (B103), while two B. atrophaeus strains (B82 and B86) showed the strongest activity towards BHET. It was established that Impranil degradation proceeded more actively at 30 °C, whereas no temperature- dependent effect was observed for bis(2-hydroxyethyl) terephthalate degradation. A total of 45 potential PETases belonging to the α/β-hydrolase superfamily were identified in the genomes of the active cultures.
