Документ Application of pcr markers for detecting 1BL.1RS wheat-rye chromosome translocations and (1 В) 1R substitutions(Одеський національний університет імені І. І. Мечникова, 2014) Motsnyi, I. Molecular-genetic and cytological analyses were carried out to detect the alien genes in original introgression stocks and to investigate their inheritance. Methods. Rye (Xremsl303, SRIR003) and wheat (XgwmJ8-1 Bs, Xgwm550-1B$, XgwmI40-1BL Xgwml53-IBL, Xgwm357-IAL, Taglut-1As) microsatellites and secalin-specific STS-marker (ω-sec-P3+ ω-.sec-P4) have been applied. Results. The (IB)1R wheat-rye chromosome substitution and 1BL.IRS translocation have been identified. The pairing between short arms of the 1 ВL .1 Rs translocation and of bread wheat chromosome 1В was observed with very low frequency (in 0.3% PMCs). Conclusions. The stocks have (1B)1R wheat-rye chromosome substitution or 1BL. 1Rs translocation. The translocation was contributed by the collection strains, derived from wheat cv. Avrora and originated from Petkus rye. The intact rye chromosome IR for the substitution was contributed by triticale (8x) cv. AD825 and originated from rye Voronezhskaya SHI. The substitution stocks were susceptible to leaf and stem rusts because of another origination of the 1R chromosome. Three major linked genes determining hairiness of the leaf upper surface (HLup). lower surface (HLlow) and leaf margin (Hlm) were revealed. Therefore, gliadin genes are an important trait/marker for breeding. Gliadin genes are localized at six main loci on the 1-st and 6-th chromosomes of each subgenome and eight minor. Two classifications of gliadins are developed on the basis of acid PAGE method: by Sozinov and Popepelya [1996] (often used in Ukraine) and by Metakovsky et al. [1995] (international). Every allelic variant in this case is a complex of linked polypeptide genes inherited as one simple Mendelian trait. Due to the complexity of APAGE method, it is possible to detect some allelic variants by PCR method during selection and breeding process. This region accounts for about a third of the annual grain production in the country. Scientific-based selection of soft wheat in Ukraine began about 80 years ago. The study of genetic diversity, the characterization of using microsatellite analysis will allow for accurate identification of varieties, estimate the level of genetic variability in the pool of Ukrainian soft wheat. Identification of the genotype of the variety is important for registration of varieties and protection of copyrights of originators. They act as growth repressors. Gibberellins (GA) promote growth by overcoming the DELLA-mediated growth restraint. A number of dominant mutations in highly conserved orthologous DELLA genes are known in various plant species. These mutations reduce plant height owing to production of more active forms of growth repressors. The aim of the current study is to estimate the genetic similarity of DELLA nucleotide and amino acid sequences stored in GenBank and to identify mutant and wild type alleles that encode DELLAs in modern Ukrainian winter bread wheat varieties. We have performed nucleotide and amino acid sequence search and alignment of the Rht-D1 wheat dwarfing gene and DELLA-encoding genes of other plant species with BLAST and MUSCLE software. A dendrogram illustrating phylogenetic relations among DELLA sequences of 35 plant species has been constructed using MEGA v5. In addition, PCR-based genotyping for the major wheat dwarfing genes Rht-B1 and Rht-D1 has been performed in wheat cultivars from different regions of Ukraine. The translocations were suggested to support a resistance to leaf rust caused by the Lr genes, being not related to them, and to determine confident resistance to stem rust. Several research works were carried out to investigate structure, distribution, evolution and relationships of Ppd-Dl gene with major agronomic traits (Beales et al., 2007; Yang et al., 2008; Guo et al., 2010). According to these works Ppd-Dl gene was sequenced and 6 alleles were revealed. There are multiple loci known as E series or eleven major genes E1 – E11 and J gene, which generally controlled time to flowering and maturity, two traits that are highly influenced by photoperiod [1, 2]. Among these loci dysfunctional alleles of maturity genes E3 and E4, which are phytochrome A genes, are involved in soybean photoperiodic insensitivity. For most genotypes of soybean varieties and lines created in different breeding centers of Ukraine, identification of alleles by allele-specific markers for loci E3, E4 was not performed. I.; Чеботарь, Сабина Витальевна; Чеботар, Сабіна ВіталіївнаInvestigation of the pleiotropic effects of GA sensitive (Rht8) and GA insensitive (Rht B1 and Rht D1) winter bread wheat dwarfing genes and the gene that determines the response of plants to photoperiod— Ppd D1—were carried out for 3 years in the southern step region of the Black Sea bank on five different genetic backgrounds. It is shown that, in addition to direct effects on plant height, GA sensitive and GA insensitive dwarf ing genes have pleiotropic effects on all studied traits except the number of fertile spikelets. Presence of the dwarfing genes in the genotype of tall forms led to the decrease of stem and ear length, and, at the same time, to the increase of ear density. The number of spikelets per spike decreased due to sterile spikelets, whereas the number of fertile spikelets did not change. There was a significant increase in the number of grains per ear as a result of increasing of spikelets in ears. The number and weight of grains did not decrease, even though the plants were charac terized by a smaller number of productive tillers. The presence of Rht8x allele on genetic background of vari ety Stepnyak resulted in a significant decrease of plants productivity. However, in combination with Ppd D1a allele, plants with Rht8x increased the potential productivity and surpassed the parental form (Rht8x Ppd D1a). This method is difficult to use, because peptides encoded by all gliadin-encoding loci are present on one lane of the gel and may overlap. Therefore, there is a need for molecular markers that would allow the determination of allelic variants of gliadins using PCR. This will help to use gliadins for marker-asissted selection, to identify allelic variants of gliadins even before obtaining grain suitable for electrophoresis of storage proteins. The data on the climate conditions of Odesa region during the last 20 years (2000-2019) were summarized and correspondence of the phenological phases of winter wheat to different periods of weather conditions during the growth in the South Steppe zone of Ukraine was analysed. It was concluded that the most negative impact of drought takes place during seed germination and early growth, at flowering and grain filling and is due to the inhibition of growth, photosynthesis, and reproductive processes.Документ Verification of near-isogenity for lines of bread weat by PCR-analysis(Одеський національний університет імені І. By using PCR-analysis we analyzed near-isogenity of lines Stepnyak 1 ranniy and Kooperatorka rannyaya, that have been created on the base of cv. Kooreratorka and cv. Stepnyak1, the degree of recovery genetics background of recurrent parental line have been shown as 100% and 98,6%, respectively.
